Noggin Is Required for Correct Guidance of Dorsal Root Ganglion Axons

MembersofthebonemorphogeneticproteinfamilyofsecretedproteinsignalshavebeenimplicatedasaxonguidancecuesforspecificneuronsinCaenorhabditiselegansandinmammals.WehaveexaminedaxonalpathfindinginmicelackingthesecretedbonemorphogeneticproteinantagonistNoggin.Wehavefounddefectsinprojectionofseveralgroupsofneurons,includingtheinitialascendingprojectionsfromthedorsalrootganglia,motoraxonsinnervatingthedistalforelimb,andcranialnerveVII.Thecaseofthedorsalrootgangliondefectisespeciallyinteresting:initialprojectionsfromthedorsalrootganglionenterthedorsalrootentryzone,asnormal,butthenprojectdirectlyintothegraymatterofthespinalcord,ratherthanturningrostrallyandcaudally.Explantexperimentssuggestthatthedefectlieswithinthespinalcordandnotthedorsalrootganglionitself.However,exogenousbonemorphogeneticproteinsareunabletoattractorrepeltheseaxons,andthespinalcordshowsonlyverysubtlealterationsindorsal–ventralpatterninNogginmutants.Wesuggestthatthedefectinprojectionintothespinalcordislikelytheresultofbonemorphogeneticproteinsdisruptingthetransductionofsomeunidentifiedrepulsivesignalfromthespinalcordgraymatter.
