【 摘 要 】

TodeterminetheroleofBonemorphogeneticprotein(BMP)signalinginmurinelimbdevelopmentinvivo,thekeratin14promoterwasusedtodriveexpressionoftheBMPantagonistNogginintransgenicmice.PhosphorylationandnucleartranslocationofSmad1/5weredramaticallyreducedinlimbsofthetransgenicanimals,confirmingtheinhibitionofBMPsignaling.Thesemicedevelopedextensivelimbsofttissuesyndactylyandpostaxialpolydactyly.Apoptosisinthedevelopinglimbnecroticzoneswasreducedwithincompleteregressionoftheinterdigitaltissue.ThepostaxialextradigitisalsoconsistentwitharoleforBMPsinregulatingapoptosis.Furthermore,therewaspersistentexpressionofFgf8,suggestingadelayintheregressionoftheAER.However,Msx1andMsx2expressionwasunchangedinthesetransgenicmice,implyingthatinductionofthesegenesisnotessentialformediatingBMP-inducedinterdigitalapoptosisinmice.TheseabnormalitieswererescuedbycoexpressingBMP4underthesamepromoterindoubletransgenicmice,suggestingthatthelimbabnormalitiesareadirecteffectofinhibitingBMPsignaling.
