ThetumorsuppressorAPCanditshomologs,firstidentifiedforaroleincoloncancer,negativelyregulateWntsignalinginbothoncogenesisandnormaldevelopment,andplayWnt-independentrolesincytoskeletalregulation.BothDrosophilaandmammalshavetwoAPCfamilymembers.WefurtherexploredthefunctionsoftheDrosophilaAPCsusingthelarvalbrainasamodel.Wefoundthatbothproteinsareexpressedinthebrain.APC2hasahighlydynamic,asymmetriclocalizationthroughthelarvalneuroblastcellcyclerelativetoknownmediatorsofembryonicneuroblastasymmetricdivisions.Adherensjunctionproteinsalsoareasymmetricallylocalizedinneuroblasts.InadditiontheyaccumulatewithAPC2andAPC1innervesformedbyaxonsoftheprogenyofeachneuroblast-ganglionmothercellcluster.APC2andAPC1localizetoverydifferentplaceswhenexpressedinthelarvalbrain:APC2localizestothecellcortexandAPC1tocentrosomesandmicrotubules.Despitethis,theyplayredundantrolesinthebrain;whileeachsinglemutantisnormal,thezygoticdoublemutanthasseverelyreducednumbersoflarvalneuroblasts.OurexperimentssuggestthatthisdoesnotresultfrommisregulationofWgsignaling,andthusmayinvolvethecytoskeletaloradhesiverolesofAPCproteins.
