【 摘 要 】
DifferentCNSregionsexhibitdifferenttemporalpatternsofoligodendrocytegenerationandmyelinogenesis.Characterizationofoligodendrocyte-type-2astrocyteprogenitorcells(hereabbreviatedasO-2A/OPCs)isolatedfromdifferentregionsindicatesthesedevelopmentalpatternsareconsistentwithpropertiesofthespecificO-2A/OPCsresidentineachregion.Markeddifferenceswereseeninself-renewalanddifferentiationcharacteristicsofO-2A/OPCsisolatedfromcortex,opticnerveandopticchiasm.Inconditionswhereopticnerve-derivedO-2A/OPCsgeneratedoligodendrocyteswithin2days,oligodendrocytesarosefromchiasm-derivedcellsafter5daysandfromcorticalO-2A/OPCsonlyafter7–10days.Thesedifferences,whichappeartobecell-intrinsic(andmayberelatedtointracellularredoxstate),weremanifestedbothinreducedpercentagesofclonesproducingoligodendrocytesandinalesserrepresentationofoligodendrocytesinindividualclones.Inaddition,responsivenessofopticnerve-,chiasm-andcortex-derivedO-2A/OPCstothyroidhormone(TH)andciliaryneurotrophicfactor(CNTF),well-characterizedinducersofoligodendrocytegeneration,wasinverselyrelatedtotheextentofself-renewalobservedinbasaldivisionconditions.Ourresultsdemonstratehithertounrecognizedcomplexitiesamongtheprecursorcellsthoughttobetheimmediateancestorsofoligodendrocytes,andsuggestthatthepropertiesofthesedifferentpopulationsmaycontributetothediversetimecoursesofmyelinationindifferentCNSregions.
