Wehavedemonstratedpreviouslythatbindingofephrin‐A1toEphAreceptorsonhumanCD4+andCD8+Tcellsstimulatesmigration.TwoEphAreceptorshavebeenreportedinTcells:EphA1attheproteinlevelandEphA4atthemRNAlevel.Inthisstudy,wewantedtoinvestigatetheexpressionprofileofthesereceptorsinTcellsubpopulationsandtotestifexpressiondifferenceswouldaffectthepotentialofcellstomigrateuponephrin‐A1binding.Wehavegeneratedananti‐EphA4mAbforexpressionanalysis.OurdatashowthatfunctionalEphA4isexpressedonthecellsurfaceofCD4+andCD8+Tcells.Inaddition,EphA4receptorexpressionisinducedafterovernightincubationinserum‐freemedium,inparticular,onCD4+CD45RO+Tcells.MigrationofCD4+Tcellsinresponsetoephrin‐A1isobservedformemorycells(CD45RO+)andmuchweakerfornaïvecells(CD45RA+).AsignalingcomplexassociatedwiththeEphA4receptorhasalsobeenisolatedandincludesEphA1,theSrcfamilykinasesFynandLck,Slp76,andVav1.Toconclude,TcellsexpressEphA1andEphA4receptors.ExpressiondifferencesofEphA4areobservedinsubpopulationsofCD4+Tcells.Thisisrelatedtothecellmigrationpotentialafterephrin‐A1binding...
