Sphingolipid‐andcholesterol‐richlipidraftmicrodomainsareimportantintheinitiationofBCRsignaling.AlthoughitisknownthatlipidraftspromotethecoclusteringofBCRandLynkinasemicroclusterswithintheBcellIS,themolecularmechanismoftherecruitmentoflipidraftsintotheBcellISisnotunderstoodcompletely.Here,wereportthatthesynapticrecruitmentoflipidraftsisdependentonthecytoskeleton‐remodelingproteins,RhoAandVav.Suchaneventisalsoefficientlyregulatedbymotorproteins,myosinIIAanddynein.Furtherevidencesuggeststhesynapticrecruitmentoflipidraftsis,byprinciple,aneventtriggeredbyBCRsignalingmoleculesandsecondmessengermolecules.BCR‐activatingcoreceptorCD19potentlyenhancessuchaneventdependingonitscytoplasmicTyr421andTyr482residues.TheenhancingfunctionoftheCD19‐PI3KmoduleinsynapticrecruitmentoflipidraftsisalsoconfirmedinhumanperipheralbloodBcells.Thus,theseresultsimproveourunderstandingofthemolecularmechanismoftherecruitmentoflipidraftmicrodomainsinBcellIS...
