Bcellsacquiremembrane‐boundcognateantigensfromthesurfaceoftheAPCsbyforminganIS,similartothatseeninTcells.Recognitionofmembrane‐boundantigensontheAPCsinitiatesadhesionofBlymphocytestotheantigen‐tetheredsurface,whichisfollowedbytheformationofradiallamellipodia‐likestructures,aprocessknownasBcellspreading.ThespreadingresponserequirestherearrangementofthesubmembraneactincytoskeletonandisregulatedmainlyviasignalstransmittedbytheBCR.Here,weshowthatcytoplasmiccalciumisaregulatorofactincytoskeletondynamicsinBlymphocytes.WefindthatBCR‐inducedcalciummobilizationisindispensibleforadhesionandspreadingofBcellsandthatPLCγandCRAC‐mediatedcalciummobilizationarecriticalregulatorsoftheseprocesses.Measuringcalciumandactindynamicsinlivecells,wefoundthatagenerationofactin‐basedmembraneprotrusionisstronglylinkedtothedynamicsofacytoplasmic‐freecalciumlevel.Finally,wedemonstratethatPLCγandCRACchannelsregulatetheactivityofactin‐severingproteincofilin,linkingBCR‐inducedcalciumsignalingtotheactindynamics...
