【 摘 要 】

Glycine soja, the wild annual ancestor of soybean, has great potential to be used as asource of genetic diversity for the crop. A Glycine soja line, PI 468916, contains aresistance QTL to soybean cyst nematode (SCN) Heterodera glycines, a devastating pestof soybean. This resistance QTL has been fine mapped in PI 468916 to a distance of147kb. Through the use of whole genome sequencing with de novo assembly we wereable to gain information about the region. In this thesis the assembly data was used alongwith fosmid sequences from both Sanger sequencing and Illumina reads to gain insightinto sequence differences between the Williams 82 reference genome and PI 468916 toidentify the gene(s) responsible for this resistance. The alignment of PI 468916 sequenceagainst the reference genome corresponding to the resistance QTL and the candidategenes present in this region, is presented. This novel resistance locus could be used tointroduce new resistance in the crop.
