【 摘 要 】
Thesinglecellgel(SCG)electrophoresisorcometassayisatechniquedetectingofmultipleclassesofDNAdamagesuchassingleanddoublestrandbreaksandoxidativebasedamageandisusedforawiderangeofapplications.ThecometassayisarelativelysensitivetestwhichenablestomeasuringDNAdamagespontaneouslyandcausedbyvariousagents.Theprincipleadvantagesofthetechniqueareapplytoanycelltypefromprokaryoticandeukaryoticorganisms,culturedcellsorcellsisolatedfromtheorganismsandit.Theotheradvantagesareitsreliability,lowcosts,easeofapplication,andtheshorttimeneededtocompleteastudy.Therefore,ithasgainedinpopularityasastandardtechniqueforevaluationofDNAdamage/repair,biomonitoringandgenotoxicitystudies.Thebasicstepsofthecometassayarepreparationofmicroscopeslideslayeredwithcellsinagarose,lysis,alkalitreatment,electrophoresisunderalkalineconditions,neutralization,DNAstaining,cometvisualizationandcometscoring.Imageanalysistoolsareavailabletoanalyzevariousparametersofthecomets.Theaimofthisreviewistogiveinformationabouttheprinciplesandapplicationsofthecometassay,asensitiveandrapidtechniqueforthemeasurementofDNAdamage.
