Backgroundandobjective:Themicrobialprofilesofstimulatedsalivasampleshavebeenshowntodifferentiatebetweenpatientswithperiodontitis,patientswithdentalcaries,andorallyhealthyindividuals.Salivawasstimulatedtoallowforeasyandrapidcollection;however,microbialcompositionmaynotreflectthemorenatural,unstimulatedstate.Thepurposeofthisstudywastovalidatewhetherstimulatedsalivaisanadequatesurrogateforunstimulatedsalivaindeterminingsalivarymicrobiomes.Design:Unstimulated(n=20)andstimulated(n=20)salivasampleswerecollectedfrom20orallyandsystemicallyhealthy,non-smokingparticipants.SalivarybacterialprofileswereanalyzedbymeansoftheHumanOralMicrobeIdentificationusingNextGenerationSequencing(HOMINGS),andstatisticalanalysiswasperformedusingMann–WhitneytestwithBenjamini–Hochberg’scorrectionformultiplecomparison,clusteranalysis,principalcomponentanalysis,andcorrespondenceanalysis.Results:Fromatotalof40salivasamples,496probetargetswereidentifiedwithameannumberoftargetspersampleof203(range:146–303),andameannumberofprobetargetsof206and200inunstimulatedandstimulatedsalivasamples,respectively(p=0.62).Basedonallstatisticalmethodsusedforthisstudy,themicrobialprofilesofunstimulatedandstimulatedsalivasamplescollectedfromthesamepersonwerenotstatisticallysignificantlydifferent.Conclusions:Analysisofbacterialsalivaryprofilesinunstimulatedandstimulatedsalivasamplescollectedfromthesameindividualshowedcomparableresults.Thus,theresultsverifythatstimulatedsalivaisanadequatesurrogateofunstimulatedsalivaformicrobiome-relatedstudies.Keywords:saliva;HOMINGS;bacteria(Published:16March2016)Citation:JournalofOralMicrobiology2016,8:30112-http://dx.doi.org/10.3402/jom.v8.30112
