【 摘 要 】
Background:Somedentalplaquefluorescesred.Thefactorsinvolvedinthisfluorescenceareyetunknown.Objective:Theaimofthisstudywastoassesssystematicallytheeffectofage,thickness,andcariogenicityontheextentofredfluorescenceproducedbyinvitromicrocosmbiofilms.Design:Theeffectsofbiofilmageandthicknessonredfluorescenceweretestedinaconstantdepthfilmfermentor(CDFF)bygrowingbiofilmsofvariablethicknessesthatreceivedaconstantsupplyofdefinedmucinmedium(DMM)andeightpulsesofsucrose/day.TheinfluenceofcariogenicityonredfluorescencewastestedbygrowingbiofilmondentindisksreceivingDMM,supplementedwiththreeoreightpulsesofsucrose/day.Thebiofilmswereanalyzedatdifferenttimepointsafterinoculation,upto24days.Emissionspectraweremeasuredusingafluorescencespectrophotometer(λexc405nm)andthebiofilmswerephotographedwithafluorescencecamera.Thecompositionofthebiofilmswasassessedusing454-pyrosequecingofthe16SrDNAgene.Results:Fromday7onward,thebiofilmsemittedincreasingintensitiesofredfluorescenceasevidencedbythecombinedredfluorescencepeaks.Theredfluorescenceintensitycorrelatedwithbiofilmthicknessbutnotinalinearway.Biofilmfluorescencealsocorrelatedwiththeimposedcariogenicity,evidencedbytheinduceddentinmineralloss.Increasingthebiofilmageorincreasingthesucrosepulsingfrequencyledtoashiftinthemicrobialcomposition.Theseshiftsincompositionwereaccompaniedbyanincreaseinredfluorescence.Conclusions:Thecurrentstudyshowsthatathicker,older,ormorecariogenicbiofilmresultsinahigherintensityofredfluorescence.Keywords:dentalplaque;fluorescence;microbiology;biofilms;microbiota;microcosm(Published:7April2016)Citation:JournalofOralMicrobiology2016,8:30346-http://dx.doi.org/10.3402/jom.v8.30346
