【 摘 要 】
TFIIHisaeukaryoticcomplexcomposedoftwosubcomplexes,theCAK(Cdkactivatingkinase)andthecore-TFIIH.Thecore-TFIIH,composedofsevensubunits(XPB,XPD,P62,P52,P44,P34,andP8),playsacrucialroleintranscriptionandrepair.Here,weperformedanextendedsequenceanalysistoestablishtheaccuratephylogeneticdistributionofthecore-TFIIHin63eukaryoticorganisms.Inspiteofthehighconservationofthesevensubunitsatthesequenceandgenomiclevels,thenon-enzymaticP8,P34,P52andP62areabsentfromoneorafewunicellularspecies.Togaininsightintotheirrespectiveroles,weundertookacomparativegenomicanalysisofthewholeproteometoidentifythegenesetssharingsimilarpresence/absencepatterns.WhilelittleinformationwasinferredforP8andP62,ourstudiesconfirmtheknownroleofP52inrepairandsuggestforthefirsttimetheimplicationofthecoreTFIIHinmRNAsplicingviaP34.
