LossofTwistfunctioninthecranialmesenchymeofthemouseembryocausesfailureofclosureofthecephalicneuraltubeandmalformationofthebranchialarches.IntheTwist−/−embryo,theexpressionofmolecularmarkersthatsignifydorsalforebraintissuesiseitherabsentorreduced,butthoseassociatedwithventraltissuesdisplayexpandeddomainsofexpression.Dorsoventralorganizationofthemid-andhindbrainandtheanterior–posteriorpatternoftheneuraltubearenotaffected.IntheTwist−/−embryo,neuralcrestcellsstrayfromthesubectodermalmigratorypathandthelate-migratingsubpopulationinvadesthecell-freezoneseparatingstreamsofcellsgoingtothefirstandsecondbranchialarches.CelltransplantationstudiesrevealthatTwistactivityisrequiredinthecranialmesenchymefordirectingthemigrationoftheneuralcrestcells,aswellasintheneuralcrestcellswithinthefirstbranchialarchtoachievecorrectlocalization.Twistisalsorequiredfortheproperdifferentiationofthefirstarchtissuesintobone,muscle,andteeth.
