Induction of a noggin-Like Gene by Ectopic DV Interaction during Planarian Regeneration

【 摘 要 】

Inpreviousstudies,wehaveshownthatdorsoventral(DV)interactionevokesnotonlyblastemaformation,butalsomorphogeneticeventssimilartothosethatoccurinregeneration.However,itisstillunclearwhatkindsofsignalmoleculesareinvolvedintheDVinteraction.ToinvestigatethesignalsystemsinvolvedintheDVinteraction,wefocusedonanoggin-likegene(Djnlg)identifiedbytheplanarianESTproject.Djnlgisthefirstnogginhomologueisolatedfromaninvertebrate.InDjNLG,thepositionsofninecysteineresidueswhichmaybeessentialfordimerformationwerewellconserved,butoverall,theaminoacidsequenceofDjNLGdidnotshowhighsimilaritytothesequencesofvertebrateNoggins.ExpressionofDjnlgwasobservedonlyintheproximalregionofthebranchstructuresinthebrainofintactplanarians,suggestingthatDjnlgmayhavearoleinpatternformationinthebrain.Interestingly,transientstrongexpressionofDjnlgwasobservedintheamputatedregionofregeneratingplanarians.Djnlg-expressingcellsweredetectedbeneaththemuscle9hafteramputationandwerethendetectedintheventralsubepidermalregionoftheblastema.TheinductionofDjnlgexpressionbyamputationwasnotaffectedbyX-rayirradiation,eventhoughthestemcellswerecompletelyeliminated,implyingtheexistenceofsignal-producingcellswhichmayprovideapositionalcuetothestemcells.InDVreversedgrafting,expressionofDjnlgwasstronglyinducedintheDVboundarybetweenthehostanddonor.TheseresultssuggestthatectopicDVinteractionmayinduceexpressionofDjnlginthepositionalcue-producingcells,andthatitmightbeinvolvedinstimulationofblastemaformationaswellasDVpatterningofthebody.
