【 摘 要 】
Duringvertebrategastrulation,aventraltodorsalgradientofbonemorphogeneticprotein(Bmp)activityestablishescellfates.Concomitantly,convergentextensionmovementsnarrowgermlayersmediolaterallywhilelengtheningthemanteroposteriorly.Here,bymeasuringmovementsofcellpopulationsinvivo,werevealthepresenceofthreedomainsofconvergentextensionmovementsinzebrafishgastrula.Ventrally,convergenceandextensionmovementsareabsent.Lateralcellpopulationsconvergeandextendatincreasingspeeduntiltheyreachthedorsaldomainwhereconvergencespeedslowsbutextensionremainsstrong.Usingdorsalizedandventralizedmutants,wedemonstratethatthesedomainsarespecifiedbytheBmpactivitygradient.InvivocellmorphologyandbehavioranalysesindicatedthatlowlevelsofBmpactivitymightpromoteextensionwithlittleconvergencebyallowingmediolateralcellelongationanddorsallybiasedintercalation.Further,singlecellmovementanalysesrevealedthatthehighventrallevelsofBmpactivitypromoteepibolicmigrationofcellsintothetailbud,increasingtailformationattheexpenseofheadandtrunk.WeshowthathighBmpactivitylimitsconvergenceandextensionbynegativelyregulatingexpressionofthewnt11(silberblick)andwnt5a(pipetail)genes,whicharerequiredforconvergentextensionbutnotcellfatespecification.Therefore,duringvertebrategastrulation,asinglegradientofBmpactivity,whichspecifiescellfates,alsoregulatesthemorphogeneticprocessofconvergentextension.
