DNAmethylationofCpGdinucleotidesbyDNAmethyltransferase1isimplicatedintheregulationoftranscriptionand,inparticular,thetranscriptionofimprintedgenes.Althoughtheoocyte-specificformofDnmt1(Dnmt1o)possessesafunctionalnuclearlocalizationsignal,itispredominantlylocalizedinthecytoplasmoftheoocyteandpreimplantationmouseembryobutundergoesatransientnuclearlocalizationduringtheeight-cellstage,whentheembryosundergocompaction.WereportherethatDnmt1oislikelyretainedinthecytoplasmbyanactiveprocess,since∼70%ofDNAmethyltransferaseactivityisretainedfollowingpermeabilizationproceduresthatresultinthereleaseof∼75%ofoocyte/embryoprotein.Treatmentoftheembryoswithagentsthatdisrupteithermicrofilamentsormicrotubuleshaslittle,ifany,effectontheretentionofDnmt1oinpermeabilizedembryos.WhileDnmt1odoesnotcolocalizewitheithermitochondriaorendoplasmicreticulum,itdoescolocalizewithannexinV,whichisknowntointeractwithDnmt1o.WealsoreportthatthetimingofnuclearentryofDnmt1oduringtheeight-cellstageisindependentofDNAreplication,transcription,andproteinsynthesis,aswellascompaction,cellcontact,andcytokinesis.Thetimeofnuclearentry,therefore,appearslinkedtothetimefollowingfertilization,whichsuggeststhatamolecularclockgovernsthetimeofnuclearimport.
