【 摘 要 】

TheWnt/β-cateninsignalingpathwayplaysmultiplerolesduringembryonicdevelopment,onlyafewofwhichhavebeenextensivelycharacterized.AlthoughdomainsofWntexpressionhavebeenidentifiedthroughoutembryogenesis,anatomicalandmolecularcharacterizationofrespondingcellshasbeenmostlyunexplored.Wehavegeneratedatransgeniczebrafishlinethatexpressesadestabilizedgreenfluorescentprotein(GFP)variantunderthecontrolofaβ-cateninresponsivepromoter.Earlyzygoticexpressionofthistransgene(TOPdGFP)mirrorsknowndomainsofWntsignalingintheembryo.LossofLef1activityresultsindecreasedreporterexpressionandposteriordefects,whilelossofTcf3(Headless,Hdl)activitydoesnotalterreporterexpression,eventhoughitresultsinlossofforebrainstructures.Inaddition,ectopicWnt1expressioncanactivatethereporter.Inolderembryos,weidentifyanumberoftransgene-expressingcellpopulationsasnovelsitesofβ-cateninsignaling.WeconcludethatourTOP-dGFPreporterlinefaithfullyillustratesdomainsofβ-cateninactivityandenablestheidentificationofresponsivecellpopulations.
