Themurineallantoiswillbecometheumbilicalarteryandveinofthechorioallantoicplacenta.Inpreviousstudies,growthanddifferentiationoftheallantoishadbeenelucidatedinwholeembryos.Inthisstudy,theextenttowhichexplantedallantoisesgrowanddifferentiateoutsideoftheconceptuswasinvestigated.Theexplantmodelwasthenusedtoelucidatecellandgrowthfactorrequirementsinallantoicdevelopment.Earlyheadfold-stagemurineallantoiseswereexplanteddirectlyontotissuecultureplasticorsuspendedintesttubes.Explantedallantoisesvascularizedwithdistal-to-proximalpolarity,theyexhibitedmanyofthesamesignalingfactorsusedbythevitellineandcardiovascularsystems,andtheycontainedatleastthreecelltypeswhoseidentity,geneexpressionprofiles,topographicalassociations,andbehaviorresembledthoseofintactallantoises.DiIlabelingfurtherrevealedthatisolatedallantoisesgrewandvascularizedintheabsenceofsignificantcellmingling,therebysupportingamodelofmesodermaldifferentiationintheallantoisthatisposition-andpossiblyage-dependent.Manipulationofallantoicexplantsbyvaryinggrowthmediademonstratedthattheallantoicendothelialcelllineage,likethatofotherembryonicvasculatures,isresponsivetoVEGF164.AlthoughVEGF164wasrequiredforbothsurvivalandproliferationofallantoicangioblasts,itwasnotsufficienttoinduceappropriateepithelializationofthesecells.Rather,otherVEGFisoformsand/ortheoutersheathofmesothelium,whosemaintenancedidnotappeartobedependentuponendothelium,mayalsoplayimportantroles.Onthebasisofthesefindings,weproposemurineallantoicexplantsasanewtoolforsheddinglightnotonlyonallantoicdevelopment,butforelucidatinguniversalmechanismsofbloodvesselformation,includingvascularsupportingcells,eitherintheintactorganismorinexistinginvitrosystems.
