【 摘 要 】
Prominentamongmoleculesthatcontrolneovascularprocessesisvascularendothelialgrowthfactor(VEGF).TheVEGFligandscompriseafamilyofwell-studiedmitogens/permeabilityfactorsthatbindcellsurfacereceptortyrosinekinases.TargetsincludeVEGFreceptor-1/Flt1andVEGFreceptor-2/Flk1.MicelackinggenesforVEGFligandorVEGFreceptor-2dieearlyingestation,makingitdifficulttodeterminetheprecisenatureofunderlyingendothelialcellularbehavior(s).Toexaminetheeffect(s)ofVEGFsignalingoncellbehaviorindetail,weconductedloss-of-functionstudiesusingavianembryos.InjectionofsolubleVEGFR-1resultsinmalformedvascularnetworksandtheabsenceoflargevessels.Inthemostseverecasesembryosexhibitedvascularatresia.Closelyassociatedwiththealteredphenotypewasaclearendothelialcellresponse—amarkeddecreaseincellprotrusiveactivity.Further,wedemonstratethatVEGFgainoffunctionstrikinglyincreasedcellprotrusiveactivity.Together,ourdatashowthatVEGF/VEGFreceptorsignalingregulatesendothelialcellprotrusiveactivity,akeydeterminantofbloodvesselmorphogenesis.WeproposethatVEGFfunctionsasaninstructivemoleculeduringdenovobloodvesselmorphogenesis.
