【 摘 要 】
Apopulationofhematopoieticprogenitorsbecomescommittedwithintheembryoproperintheflooroftheaorta(P-Sp/AGMinthemouse).Inbirds,thisfirstaspectofintraembryonichematopoiesisisprominentduringembryonicday3(E3)asendothelium-associated“intra-aorticclusters.â€BetweenE6andE8,diffusehematopoiesisthenoccursas“para-aorticfociâ€locatedinthedorsalmesenteryventraltotheaorta.Thesefociarenotassociatedwithendothelium.Whetherthesetwohematopoieticcellpopulationsarisefromdistinctorcommonprogenitorsisnotknown.Wecouldrecentlytracebacktheoriginofintra-aorticclustersintheavianembryobylabelingaorticendothelialcells(EC)invivowithacetylatedlow-densitylipoproteins.Thisapproachestablishedthederivationofearlyintraembryonichemopoieticcellsfromtheendothelium,butdidnotindicatehowlongduringontogenysucharelationshipmayexist,sincetheprogenyofEClabeledatE2couldbetracedfor1–2daysatmost.Herewereportthat,whenE2aorticECswereinfectedpriortotheformationofintra-aorticclusterswithanonreplicativeLacZ-bearingretroviralvector,numerouscellswerelabeledinthepara-aorticfociatE6.Incontrast,whentheretroviralvectorwasinoculatedatE4ratherthanE2,thatis,afterthedisappearanceofintra-aorticclusters,nocellsinthepara-aorticfociwerelabeled.Takentogether,ourresultsdemonstratethatECsfromtheaorticfloorseedthetwoaspectsofaorta-associatedhemopoiesisandthattheseECswithhemangioblasticpotentialarepresentonlytransientlyintheaorta.
