Anexperimentalparadigmwasdevisedtoremovetheretinalbasallaminafordefinedperiodsofdevelopment:thebasallaminawasdissolvedbyinjectingcollagenaseintothevitreousofembryonicchickeyes,anditsregenerationwasinducedbyachasewithmouselaminin-1andα2-macroglobulin.Thelaminin-1wasessentialinreconstitutinganewbasallaminaandcouldnotbereplacedbylaminin-2orcollagenIV,whereasthemacroglobulinservedasacollagenaseinhibitorthatdidnotdirectlycontributetobasallaminaregeneration.Theregenerationoccurredwithin6hafterthelaminin-1chasebyformingamorphologicallycompletebasallaminathatincludedallknownbasallaminaproteinsfromchickembryos,suchaslaminin-1,nidogen-1,collagensIVandXVIII,perlecan,andagrin.Thetemporaryabsenceofthebasallaminahaddramaticeffectsonretinalhistogenesis,suchasanirreversibleretractionoftheendfeetoftheneuroepithelialcellsfromthevitrealsurfaceoftheretina,theformationofadisorganizedganglioncelllayerwithanincreaseinganglioncellsby30%,andtheappearanceofmultipleretinalectopias.Finally,basallaminaregenerationwasassociatedwithaberrantaxonsfailingtocorrectlyentertheopticnerve.Thepresentdatademonstratethatatransientdisruptionofthebasallaminaleadstodramaticandprobablyirreversibleaberrationsinthehistogenesisinthedevelopingcentralnervoussystem.
