GenBankentriesformouseIl33revealtheexistenceoftwotranscripts,Il33aandIl33b,withdifferent5′UTRsbutcodingforthesameprotein.Weinvestigatedexpressionofthesetranscriptsindifferentmouseorgansandcelltypesinbasalandinflammatoryconditions.Il33aandIl33bmRNAsstartwithdifferentnoncodingfirstexons,transcribedfromdifferentpromoterregions,whichbothcontainaconsensusTATA‐likesequence.ConstitutiveIl33amRNAexpressionwasdetectedinmousestomach,lung,spleen,andbrain,whereasbasalIl33bmRNAexpressionwasobservedonlyinthestomach.ExpressionofbothtranscriptsincreasedaftersystemicLPSadministration.Invitro,weobservedhighconstitutiveexpressionofIl33transcriptsinMEFs.ConstitutiveIl33amRNAexpressionwasobservedalsoinBMDCs,whereitwaspreferentiallyincreasedinresponsetopoly(I:C),whereasLPSincreasedlevelsofIl33aandIl33bmRNA.Incontrast,BMMsandRaw264.7cellsdidnotexpressIl33mRNAconstitutively,andLPSstimulationselectivelyinducedexpressionofIl33bmRNAinthesecells.OurdataindicatethattheIl33geneisexpressedfromtwoalternativepromotersinthemouseandthattherelativeexpressionofIl33aandIl33btranscriptsiscelltype‐andstimulus‐dependent...
