ASS1isacytosolicenzymethatplaysaroleintheconversionofcitrullinetoarginine.Inhumanandmousetissues,ASS1proteinisfoundinseveralcomponentsoftheimmunesystem,includingthethymusandTcells.However,theroleofASS1inthesetissuesremainstobedefined.ConsiderableattentionhasbeenfocusedrecentlyontheroleofmetabolisminTcelldifferentiationandfunction.BasedontheexpressionofASS1intheimmunesystem,wehypothesizedthatASS1deficiencywouldresultinTcelldefects.Toevaluatethisquestion,wecharacterizedimmunefunctioninhypomorphicfold/foldmice.AnalysisofsplenicTcellsbyflowcytometryshowedamarkedreductioninTcellnumberswithnormalexpressionofactivationsurfacemarkers.GenetherapycorrectionofliverASS1toenhancesurvivalresultedinapartialrecoveryofsplenicTcellsforcharacterization.InvitroandinvivostudiesdemonstratedthepersistenceoftheASS1enzymedefectinTcellsandabnormalTcelldifferentiationandfunction.Overall,ourworksuggeststhatASS1playsaroleinTcellfunction,anddeficiencyproducesprimaryimmunedysfunction.Inaddition,thesedatasuggestthatpatientswithASS1deficiency(citrullinemiatypeI)mayhaveTcelldysfunction...
