Multiplepathologicconditions,includinghemorrhage,tumorangiogenesis,andischemia‐reperfusionevents,willresultinhypoxiaandsubsequentreperfusion.Previousstudieshaveanalyzedthelipidchangeswithinwholetissuesandindicatedthatischemia‐reperfusionalteredtissueandcellularphospholipids.Usinganinvitrocellculturemodelofhypoxiaandreoxygenation,weexaminedtheendotheliallipidchanges.Wehypothesizedthatphospholipidscramblase1,aproteinthatregulatesbilayerasymmetry,isinvolvedinalteringthephospholipidsofendothelialcellsduringhypoxia,acomponentofischemia,leadingtoβ2‐glycoproteinIandIgMbindingandsubsequentlipid‐mediated,inflammatoryresponses.Wehavecompletedthefirstcomprehensivestudyofsteady‐statephospholipidscramblase1mRNAlevels,proteinexpression,andactivityunderconditionsofhypoxiaandreoxygenation.Phospholipidscramblase1regulatesphosphatidylserineexposureinresponsetooxygenstress,leadingtoβ2‐glycoproteinIandIgMbindingandlipid‐mediated,inflammatoryresponses...
