InfectionbyHIVisassociatedwiththeexpansionofmonocytesexpressingCD16antigens,butthesignificanceofthisinHIVpathogenesisislargelyunknown.Inrhesusmacaques,atleastthreesubpopulationsofbloodmonocyteswereidentifiedbasedontheirexpressionofCD14andCD16:CD14highCD16−,CD14highCD16low,andCD14lowCD16high.Thephenotypesandfunctionsofthesesubpopulations,includingCD16+monocytes,wereinvestigatedinnormal,uninfectedrhesusmacaquesandmacaquesthatwereinfectedwithSIVorchimericSHIV.ToassesswhetherthesedifferentmonocytesubpopulationsexpandorcontractinAIDSpathogenesis,weconductedacross‐sectionalstudyof54SIV‐orSHIV‐infectedmacaquesand48uninfectedcontrols.Theabsolutenumbersofmonocytepopulationswereexaminedinacutelyinfectedanimals,chronicallyinfectedanimalswithnodetectableplasmavirusRNA,chronicallyinfectedanimalswithdetectableplasmavirusRNA,andanimalsthatdiedwithAIDS.TheabsolutenumbersofCD14highCD16lowandCD14lowCD16highmonocyteswereelevatedsignificantlyinacutelyinfectedanimalsandchronicallyinfectedanimalswithdetectableplasmavirusRNAcomparedwithuninfectedcontrols.Moreover,asignificant,positivecorrelationwasevidentbetweenthenumberofCD14highCD16loworCD14lowCD16highmonocytesandplasmaviralloadintheinfectedcohort.Thesedatashowthedynamicchangesofbloodmonocytes,mostnotably,CD14highCD16lowmonocytesduringlentiviralinfection,whicharespecifictodiseasestage...
