【 摘 要 】
EffectorandmemoryCD4andCD8Tcellresponsesarecriticalforthecontrolofmanyintracellularpathogens.Thedevelopmentofthesepopulationsisgovernedbytranscriptionfactorsthatmolecularlycontroltheirdifferentiation,function,andmaintenance.TwotranscriptionfactorsknowntobeinvolvedintheseprocessesareTbetandSTAT4.AlthoughTbethasbeenshowntoregulateCD8TcellfatedecisionsandeffectorCD4Tcellchoices,thecontributionofSTAT4islesswellunderstood.Toaddressthis,weexaminedtheimpactofSTAT4onTcellresponsesinthepresenceorabsenceofTbet,followingLCMVinfectionbyusingmicelackingTbet,STAT4,orbothtranscriptionfactors.STAT4wasnotrequiredforTbetorEomesexpression;however,virus‐specificeffectorCD8Tcellsareskewedtowardamemory‐precursorphenotypeintheabsenceofSTAT4.ThisalteredproportionofmemoryprecursorsdidnotresultinanincreaseinmemoryCD8Tcellsaftertheresolutionoftheinfection.Wealsodemonstratethatvirus‐specificeffectorandmemoryCD4TcellsformedindependentlyofTbetandSTAT4,althoughaslightreductioninthenumberofantigen‐specificCD4Tcellswasapparentinmicelackingbothtranscriptionfactors.Collectively,wehavediscovereddistinctrolesforTbetandSTAT4inshapingthephenotypeandfunctionofvirus‐specificTcellresponses...
