Alum‐basedadjuvantsfacilitatevaccine‐drivenhumoralimmunity,buttheirmechanismofactionremainspoorlyunderstood.Herein,wereportthatlackoftypeIINKTcellsisassociatedwithintact,matureBcellsbutdampenedhumoralimmunityfollowingimmunizationwithAlum‐adsorbedT‐dependentantigen.TypeIINKTcellsfacilitatedproductionofIL‐4,IL‐5,IL‐10,IL‐13,andantibodybyLNandsplenocyteculturesfollowingAlum/antigenadministrationinvivoandantigenrestimulationinvitro.AdditionofIL‐4andIL‐5totypeIINKT‐deficientculturesrestoredinvitroantibodyproduction.IntracellularstainingrevealedthatAlum‐primedtypeIINKTcellscoordinatedIL‐4secretionbyTcells.AlumdidnotsignificantlyaffectCD1dexpressioninvivo,butadditionofCD1d‐blockingmAbdiminishedcytokineproductionandinvitroantibodyproduction.TypeIINKTcellsthereforefunctionaspartoftheAlum‐sensingapparatusandinaCD1d‐dependentmanner,facilitateTH2‐drivenhumoralimmunity.ThismayhaveimportantconsequencesforunderstandingthemechanismofactionofAlum‐containingvaccines...
