【 摘 要 】
MDSCsareaheterogeneousgroupofmyeloidcellsthatsuppressTcellactivityincancerandautoimmunedisease.TheeffectofMDSCsonBcellfunctionisnotclear.UsingtheCIAmodelofautoimmunedisease,wefoundanincreaseinM‐MDSCsintheperipheryofWTmicewithCIAcomparedwithnai¨vemice.TheseMDSCswereabsentfromtheperipheryofCCR2−/−micethatdevelopedexacerbateddisease.M‐MDSCs,isolatedfromimmunizedmice,inhibitedautologousCD4+Tcellproliferation.TheM‐MDSC‐mediatedsuppressionofTcellproliferationwasNOandIFN‐γdependentbutIL‐17independent.Furthermore,wedemonstratedforthefirsttimethatM‐MDSCsfromCIAmicealsoinhibitedautologousBcellproliferationandantibodyproduction.ThesuppressionofBcellsbyM‐MDSCswasdependentontheproductionofNOandPGE2andrequiredcell–cellcontact.AdministrationofM‐MDSCsrescuedCCR2−/−micefromtheexacerbatedCIAphenotypeandameliorateddiseaseinWTmice.Furthermore,adoptivetransferofM‐MDSCsreducedautoantibodyproductionbyCCR2−/−andWTmice.Insummary,M‐MDSCsinhibitTcellandBcellfunctioninCIAandmayserveasatherapeuticapproachinthetreatmentofautoimmunearthritis...
