Recentstudieshaveshownthatthehumancathelicidin,LL‐37,hasantiviralactivityagainstIAVinvitroandinvivo.NeutrophilsareimportantcellularcomponentsoftheinitialinnateresponsetoIAVinfection.Inadditiontoitsdirectantimicrobialactivities,LL‐37hasimportantimmunomodulatoryeffects.Inthisstudy,weexplorehowLL‐37affectsinteractionsofIAVwithhumanneutrophils.LL‐37didnotalterneutrophiluptakeofIAVbutsignificantlyincreasedneutrophilH2O2responsestothevirus.IAVstimulatedproductionofNETsinvitro,andthisresponsewasincreasedbypreincubatingtheviruswithLL‐37.NADPH‐oxidaseblockadedidnotreduceIAV‐inducedNETformationortheincreasedNETresponsestimulatedbyLL‐37+IAV.TheincreasedrespiratoryburstandNETresponseswere,however,inhibitedbypreincubatingcellswithaformylpeptidereceptorblocker,indicatingthatLL‐37engagesthesereceptorswhencomplexedwithIAV.ResponsestoIAValonewerenotinhibitedbyformylpeptidereceptorblockade.IthasbeenreportedthatLL‐37reducesproinflammatorycytokineresponsesduringIAVinfectioninvivo.WenowshowthatIAValonepotentiatedreleaseofIL‐8fromneutrophils,andpreincubationwithLL‐37reducedIAV‐stimulatedIL‐8release.TheseresultsconfirmthatLL‐37modulateshumanneutrophilresponsestoIAVinadistinctivemannerandcouldhaveimportantbearingontheprotectiveeffectsofLL‐37duringIAVinfectioninvivo...
