CD4+CD25+Foxp3+TregshaveadiminishedcapacitytoactivatethePI3K/Aktpathway.AlthoughbluntedAktactivityisnecessarytomaintainTregfunction,theconsequencesofthisalteredsignalingareunclear.Glut1isacell‐surfacereceptorresponsibleforfacilitatingglucosetransportacrossplasmamembranes,whoseexpressionistightlycoupledtocostimulatorysignalsandAktphosphorylation.FreshlyisolatedhumanTregswereunabletoup‐regulateGlut1inresponsetoTCRandcostimulatorysignalscomparedwithTconv.Consequently,theabilityofTregstouseglucosewasalsoreduced.IntroductionofFoxp3intoTconvinhibitedAktactivationandGlut1expression,indicatingthatFoxp3canregulateGlut1.Finally,pharmacologicactivationofAktinTregscaninduceGlut1,overcomingtheeffectsofFoxp3.Together,theseresultsillustratethemolecularbasisbehinddifferentialglucosemetabolisminTregs...
