【 摘 要 】
Thenaturalcompound2,3‐BTDhasdiversephysiologicaleffectsinarangeoforganisms,includingactingasadetoxifyingproductofliveralcoholmetabolisminhumansandamelioratingendotoxin‐inducedacutelunginjuryinrats.Inthisstudy,werevealthat2,3‐BTDenhancesNKcellcytotoxicactivityinhumanpNKcellsandNK92cells.TreatmentofNKcellswith2,3‐BTDincreasedperforinexpressioninadose‐dependentmanner.ThiswasaccompaniedbyelevatedJNKandERK1/2MAPKactivitiesandenhancedexpressionofNKG2D/NCRs,upstreamsignalingmoleculesoftheMAPKpathways.The2,3‐BTDeffectwasinhibitedbypretreatmentwithinhibitorsofJNK(SP)orERK1/2(PD)orbydepletingNKG2D/NCRsorJNK1orERK2withsiRNA.Theseresultsindicatethat2,3‐BTDactivatesNKcellcytotoxicitybyNKG2D/NCRpathwaysandrepresentthefirstreportofthe2,3‐BTDeffectonactivationofinnateimmunitycells...
