IFN‐α/βwasfirstdescribedasapotentinhibitorofviralreplication,butitisnowappreciatedthatIFNsignalingplaysapleiotropicroleinregulatingperipheralTcellfunctions.Recently,IFN‐α/βwasshowntoblockhumanTh2developmentbysuppressingthetranscriptionfactorGATA3.ThiseffectisconsistentwiththeroleforIFN‐α/βinsuppressingallergicinflammatoryprocessesbyblockinggranulocyteactivationandIL‐4‐mediatedBcellisotypeswitchingtoIgE.WiththeconsiderationofrecentstudiesdemonstratingadefectinIFN‐α/βsecretioninDCsandepithelialcellsfromindividualswithsevereatopicdiseases,thereisanapparentreciprocalnegativeregulatoryloopinatopicindividuals,wherebythelackofIFN‐α/βsecretionbyinnatecellscontributestothedevelopmentofallergicTh2cells.IsitpossibletoovercometheseeventsbytreatingwithIFN‐α/βorbyinducingitssecretioninvivo?Insupportofthisapproach,casestudieshavedocumentedthetherapeuticpotentialofIFN‐α/βintreatingsteroid‐resistantallergicasthmaandotheratopicdiseases.Additionally,individualswithasthmawhoareinfectedwithHCVandrespondtoIFNtherapyshowedareductioninsymptomsandseverityofasthmaattacks.Thesefindingssupportamodel,wherebyallergicandantiviralresponsesareabletocross‐regulateeachother,asIgERcross‐linkingofpDCspreventsIFN‐α/βproductioninresponsetoviralinfection.Theclinicalimportanceofupper‐respiratoryvirusesinthecontextofallergicasthmasupportstheneedtounderstandhowthesepathwaysintersectandtoidentifypotentialtherapeutictargets...
