Phagocytosisisanancientcellularprocessthatplaysanimportantroleinhostdefense.InDrosophilamelanogasterphagocytic,macrophage‐likehemocytesrecognizeandingestmicrobes.WeperformedanRNAi‐basedinvitroscreenintheDrosophilahemocytecelllineS2andidentifiedAbi,cpa,cofilinregulator14‐3‐3ζ,tlk,CG2765,andCG15609asmediatorsofbacterialphagocytosis.Oftheseidentifiedgenes,14‐3‐3ζhadanevolutionarilyconservedroleinphagocytosis:bacterialphagocytosiswascompromisedwhen14‐3‐3ζwastargetedwithRNAiinprimaryDrosophilahemocytesandwhentheorthologousgenesYwhabandYwhazweresilencedinzebrafishandmouseRAW264.7cells,respectively.InDrosophilaandzebrafishinfectionmodels,14‐3‐3ζwasrequiredforresistanceagainstStaphylococcusaureus.Weconcludethat14‐3‐3ζisessentialforphagocytosisandmicrobialresistanceininsectsandvertebrates...
