Asweage,thecompositionofourperipheralleukocyteschangesdramatically.Manyofthesealterationscontributetothegeneralimmunedysfunctionthatburdenstheelderly,whichinturn,contributestoincreasedsusceptibilitytodisease.MDSCsrepresentaheterogeneouspopulationofimmunosuppressiveleukocytesthatareelevatedintheperipheralbloodofcancerpatients.Giventherelationbetweencancerincidenceandage,thisstudyexaminedthefrequencyofperipheralbloodCD33(+)HLA‐DR(−)MDSCsacrossthreecohorts:healthyadults(19–59yearsold),community‐dwellingseniors(61–76yearsold),andfrailelderly(67–99yearsold).ThisanalysisisthefirsttodemonstratethatMDSCsandspecificallytheCD11b(+)CD15(+)MDSCsubsetareincreasedwithage.ProinflammatorycytokinesthatarerequiredforthedifferentiationofMDSCs(e.g.,TNF‐α,IL‐6,andIL‐1β)weresimilarlyfoundtobeincreasedintheserumofthefrailelderly.Furthermore,theproportionofMDSCsandtheCD11b(+)CD15(+)subsetwerefoundtobeelevatedsignificantlyinelderlydonorswithahistoryofcancer.Thisage‐relatedelevationinthefrequencyofMDSCsmaycontributetotheincreasedcancerincidencethatoccurswithage.FurtherinvestigationintothefunctionalconsequencesofelevatedMDSCswillprovidevaluableinsightintotheprogressionofage‐relatedpathologies...
