ByinteractingwithCD26ontheCD4+TcellsurfaceandwiththeAdoRA2BontheDCsurface,ADAtriggersacostimulatorysignalforhumanTcells.TheaimofthisstudywastoknowwhetherADA‐mediatedcostimulationplaysaroleinthedifferentiationofTcells.TheresultsshowthatirrespectiveofitsenzymaticactivityanddependentonTNF‐α,IFN‐γ,andIL‐6action,ADAenhancedthedifferentiationofCD4+CD45RA+CD45RO–naïveTcellstowardCD4+CD25+CD45RO+TeffsandCD4+CD45RA–CD45RO+memoryTcells.Furthermore,ADApotentiatedgenerationofCD4+CD25highFoxp3+TregsbyamechanismthatseemstobemainlydependentontheenzymaticactivityofADA.Interestingly,anADA‐mediatedincreaseonTeff,memoryTcell,andTreggenerationoccurred,notonlyincoculturesfromhealthyindividualsbutalsofromHIV‐infectedpatients.ThesedatasuggestthatADAisarelevantmodulatorofCD4+Tcelldifferentiation,evenincellsfromimmunologicallycompromisedindividuals...
