Thetestisisasiteofimmuneprivilegeinrodents,andthereisevidencethatTcellresponsesarealsosuppressedintheprimatetestis.LocalimmunosuppressionisapotentialmechanismforHIVpersistenceintissuereservoirsthatfewstudieshaveexamined.Theresponseofthepig‐tailedmacaquetestistoSIVmac239infectionwascharacterizedtotestthispossibility.Testesweresurgicallyremovedduringearly‐chronic(10wk)andlate‐chronic(24–30wk)SIVinfectionin4animalsandcomparedwiththosefrom7uninfectedanimals.SIVinfectioncausedonlyminordisruptiontotheseminiferousepitheliumwithoutmarkedevidenceofinflammationorconsistentchangesintotalintratesticularleukocytenumbers.InfectionalsoledtoanincreaseintherelativeproportionoftesticulareffectormemoryCD8+TcellnumbersandacorrespondingreductionincentralmemoryCD4+Tcells.Adecreaseintherelativeproportionofresident‐typeCD163+macrophagesandDCswasalsoobserved.SIV‐specificCD8+Tcellsweredetectableinthetestis,10–11wkafterinfectionbystainingwithSIVGag‐specificorTat‐specificMHC‐Itetramers.However,testicularCD8+Tcellsfromtheinfectedanimalshadsuppressedcytokineresponsestomitogenactivation.TheseresultssupportthepossibilitythatlocalimmunosuppressioninthetestismayberestrictingtheabilityofTcellstorespondtoSIVorHIVinfection.LocalimmunosuppressioninthetestismaybeanunderexploredmechanismallowingHIVpersistence...
