Thecurrentstudyreportsaflowcytometry‐basedprotocolfortheprospectivepurificationofhumanBMpopulationsrepresentingsixsuccessivestagesofterminalneutrophildifferentiation,includingearlypromyelocytesandlatepromyelocytes,myelocytes,metamyelocytes,bandcells,andPMNneutrophilicgranulocytes.Validationexperimentsrevealedahighpurityofeachbonemarrowpopulationandbiologicalmeaningfulexpressionprofilesformarkergenesofneutrophildifferentiationatahithertounprecedentedresolution.Hence,thepresentprotocolshouldbeusefulforstudyingneutrophildifferentiationinvivointhehumansettingandconstitutesanimportantalternativetomodelsthatarebasedoninvitrodifferentiationofmyeloidcelllinesandHPCs...
