【 摘 要 】
TheobjectiveofBiologicalDosimetryisitsapplicabilityinradioprotection,quantifyingthedosereceivedinindividualswhohavebeenexposedtoionizingradiation.Forthisraison,theanalysisofthefrequencyofchromosomeaberrationspresentinfirstdivisionmetaphasesfromstimulatedlymphocytesisacceptedasbeingthereliable.However,inproliferatingcellsthereareseveralcheckpointsthatpreventdamagedcellstopassthrough,andheavilydamagedcellsshowanimpossibilitytoreachmitosis.ConcretelythechemicallyinducedprematurechromosomecondensationbyCalyculin-AincombinationwithColcemiditispossibletoscoreinthesameslidecellsthatwereininterphaseandinmetaphase.TheaimistodetermineifCalyculin-Ahasanyeffectinthedicentricyieldobservedinmetaphasecells.Peripheralbloodsampleswereirradiatedat1,4,and10Gyofγ-raysandculturedfor48hours.Colcemidwasadded24or2hoursbeforeharvesting,andforcomparisonscultureswerealsotreatedwithofCalyculin-Aatthelasthour.ForthethreedosestheproportionofMcellswasalwayshigherafter24hoursinpresenceofColcemidthanafter2hours.Whenthefrequenciesofdicentricswerecompared,therewerenomajordifferencesbetweencultureswhereonlyColcemidwasused(2or24hours)andthosewheretheCalyculin-Atreatmentwasalsodone.TheresultsindicatethatwhenPCC-CAisusedifmetaphasecellsareconsideredsimilardose-effectcurveswillbeobtainedthanthoseobtainedusingtheconventionalmitoticarrestwithColcemid.
