Methods.FromOctober2006toMay2010146molecularHLA-typingswereperformedintheRegionalChildren’sHospitalNo.1for72patientsand74potentialrelateddonors.Respectively,142and24sampleswereanalyzedbyPCRwithsequence-specificprimersandsequence-basedHLA-typing.Bothmethodswereappliedin20cases.Results.Among84examinedsiblings,11completelyidenticalpairs,39haploidenticalcases,and23mismatchedcaseswerefound.Forty-sevendonorrequestsearchesweresenttolocalstemcelldonorregistries,and11tocordbloodbanksthroughouttheRussianFederation.Resultsareshowninthetablebelow.Low-resolutionDNA-typingidentified15matcheddonors(9/10and10/10matches).High-resolutionDNAtyping,performedattheMolecularbiologylaboratoryofRegionalChildrenHospitalNo.1,allowedustodetect4matched(10/10)unrelateddonors,22HLA-identicalsiblings,andoneuniquecaseofamother-daughtermatchedpair.In9detectedmatchedpairs(5relatedand4unrelated)DNAsamplesweresentforreferenceHLA-typingtoanEFI-accreditedHLA-laboratory(Austria).Nodiscrepancieswerefound.Conclusions.Ourpreliminarydatashowsthatforwide-spreadHLA-genotypesmatcheddonorscanbefoundevenwithinrelativelysmalllocalregistries.LocalregisterNumberofrequestsResultsoflow-resolutionHLA-typing10/10matches9/10matcheslt;9/10matches6/6matches5/6matcheslt;5/6matchesStemcelldonorregistriesStemcelldonorregistry,Chelyabinsk28597–––RussianCenterofimmunologicaltissuetyping,St.Petersburg11–––––47Regionallaboratoryofimmunologicaltissuetyping,Pervouralsk71––––31Karelianregistryofunrelatedhematopoieticstemcelldonors,Petrozavodsk––––––1CordbloodbanksStemcellbank,Moscow4–––––8Clinicalcenterofcelltechnology,Samara7–––23684Total58697236170
