【 摘 要 】
Background:Duetoexistingperspectivesontheclinicalusageofbonemarrowmesenchymalstemcell(MSC)populations,themostcurrenttasksaredevelopmentoftheirisolation,expansion,andinfusionmethods.Presently,however,inmostprotocolsforMSCexpansion,theculturemediumincludestheadditionoffetalcowserum(FCS),whichisapotentialsourceofxenogenicantigensaswellinfectioncontamination.Humanplateletlysate(HPL)fromplateletenrichedplasmacouldbeconsideredasanalternativetoFCS.Objectives:TheaimofthestudywasthedevelopmentofHPLextractiontechniques,andevaluationofitsbiologicalactivityandpossibleuseasanalternativetoFCS.Methods:WeinvestigatedtheproliferativeactivityofMSCsinprimarycultureandduringlong-termcultivation.Theimmunophenotype,geneexpressionprofile,differentiationability,andhematopoiesissupportfunctionwerecomparedwithFCS-supplementedandserum-freemedium.Results:InassessingMSCs'colonyformationinprimaryculturethenumberofcolony-formingunitsoffibroblasts(CFU-f)washigherwhenHPLwasusedincomparisonwithFCS.DifferencesinMSCs'proliferationcapacitywasnotobservedduringlong-termcultivation.AlowernumberofCD14+cellsandreducedlevelofNGF,osteopontin,ITGA7,andNTF3geneexpressionwasidentifiedwhenHPLwasused.TheabilityofMSCs'osteogenicdifferentiationwashigherinHPL-supplementedmedium.Conclusion:DuringexpansionofMSCsusingHPLtheirdifferentiationabilitywaschangedwithaccompaniedgeneexpressionprofilechangesandwithproliferationabilitythatwascomparabletoFCS.
