Background:Specificpathogenicbacteriahavebeenimplicatedinrecurrentaphthousstomatitis(RAS),achronicinflammatoryconditioncharacterisedbyulcerationsintheoralmucosa.However,theaetiologybehindthisconditionstillremainsunclear.Objective:ThebuccalmicrobiotaofpatientswithRASwascomparedtothatofcontrolsubjectstoinvestigateitspotentialroleforthiscondition.Design:Buccalswabswereobtainedfromnon-ulcerativeareasof60patients,ofwhom42patientshadlesionsatthetimeofsampling,and60healthyage-andgender-matchedcontrols.BacterialDNAwasextractedandanalysedbyTerminal-RestrictionFragmentLengthPolymorphism,usingenzymaticdigestionofthepolymerasechainreaction-amplified16SrRNAgene,yieldingaseriesofpeaks,eachrepresentingabacterialtaxon.Results:Twopeaks,60and489,weremoreprevalentinpatientswithRASthancontrols.Conversely,peaks58and490werelesscommoninpatientsthancontrols.Whenthepatientsweredividedintosubgroups,wefoundthattheobserveddifferencesinpeak-patternwererelatedtothepresenceoflesionsduringsampling.Conclusions:Themicrobiotaofthenon-inflamedbuccalmucosadifferedbetweenpatientsandcontrols.Thedifferencesweremostpronouncedinpatientswhopresentedwithlesionsduringsampling,suggestingthatadisturbanceinthenormalbuccalmicrobiotatriggersthepresenceoflesionsorthatpresenceoflesionsaltersthemicrobiota.Keywords:oralmucosa;oralmucosaldisease;oralbacteria;Terminal-RestrictionFragmentLengthPolymorphism;oralulceration;oralcavity(Published:29October2014)Citation:JournalofOralMicrobiology2014,6:25739-http://dx.doi.org/10.3402/jom.v6.25739Toaccessthesupplementarymaterialforthisarticle,pleaseseeSupplementaryfilesunderArticleToolsonline
