【 摘 要 】
Atypicalantipsychotic(AAP)drugsarethepreferredchoiceoftreatmentforschizophreniapatients.PatientswhodonotshowfavorableresponsetoAAPmonotherapyaresubjectedtorandomprolongedtherapeutictreatmentwithAAPmultitherapy,typicalantipsychoticsoracombinationofboth.Therefore,prioridentificationofpatients'responsetodrugscanbeanimportantstepinprovidingefficaciousandsafetherapeutictreatment.Wethusattemptedtoelucidateageneticsignaturewhichcouldpredictpatients'responsetoAAPmonotherapy.Ourlogisticregressionanalysesindicatedtheprobabilitythat76%patientscarryingcombinationoffourSNPswillnotshowfavorableresponsetoAAPtherapy.Therobustnessofthispredictionmodelwasassessedusingrepeated10-foldcrossvalidationmethod,andtheresultsacrossn-foldcross-validations(meanaccuracy#xA0;=#xA0;71.91%;95%CI#xA0;=#xA0;71.47ndash;72.35)suggesthighaccuracyandreliabilityofthepredictionmodel.Furthervalidationsoftheseresultsinlargesamplesetsarelikelytoestablishtheirclinicalapplicability.
