【 摘 要 】
Manymolecularmechanismscontributetothedevelopmentofdoxorubicinresistanceanddifferentcancerscanexpresswideanddiversearraysofdrug-resistancegenes.TheaimofthisstudywastoidentifythechangesingeneexpressionassociatedwiththedevelopmentofdoxorubicinresistanceinMCF7breastcancercellline.ThedoxorubicinresistantMCF7celllinewasdevelopedbystepwiseselectionofMCF7cellsandwastestedusingtheMTTassay.Thealterationsingeneexpressionwereexaminedusingthereal-timebasedPCRarray.Thefindingsshowedanup-regulationofmanyphaseI/IImetabolizinggenes,specifically,theCYP1A1andtheCYP1A2thatwereup-regulatedby206-and96-foldrespectively.Drugeffluxpumpgeneswerealsoup-regulatedprofoundly.TOP2Awasstronglydown-regulatedby202-fold.Manyotherchangeswereobservedingenescrucialforcellcycle,apoptosisandDNArepair.Thefindingsofthisprojectimplythatthedevelopmentofdoxorubicinresistanceisamulti-factorialprocess.
