【 摘 要 】
PretermbirthintheUnitedStatesisnow12%.Multiplegenes,genenetworks,andvariantshavebeenassociatedwiththisdisease.Usingacustomdatabaseforpretermbirth(dbPTB)witharefinedsetofgenesextensivelycuratedfromliteratureandbiologicaldatabases,weanalyzedGWASofpretermbirthforcompletegenotypedataonnearly2000pretermandtermmothers.Weusedboththecuratedgenesandagenome-wideapproachtocarryoutapathway-basedanalysis.Therewere19significantpathways,whichwithstoodFDRcorrectionformultipletestingthatwereidentifiedusingboththecuratedgenesandthegenome-wideapproach.Theanalysisbasedonthecuratedgeneswasmoresignificantthangenome-widein15outof19pathways.Thisapproachdemonstratestheuseofavalidatedsetofgenes,intheanalysisofotherwiseunsuccessfulGWASdata,toidentifygenendash;geneinteractionsinawaythatenhancesstatisticalpoweranddiscovery.
