Smallnon-codingregulatoryRNAs(sRNAs)playimportantrolesinregulatinggeneexpressionatthepost-transcriptionallevelandoftenrequiretheRNAchaperoneHfq.ThehumanopportunisticpathogenBurkholderiacenocepaciaJ2315encodestwodistinctRNAchaperones,HfqandHfq2.Thepresentworkdescribestheexperimentalidentificationandvalidationof24sRNAsfromB.cenocepaciaJ2315,basedontheco-purificationofsRNAswiththebacteriumHfqprotein,followedbyconversionintocDNA,cloning,computationalanalysisofsequencesandvalidationbyNorthernblotanalysis.ThesRNAsherereportedescapedidentificationbypreviousstudiesbasedontranscriptomicsorbioinformaticanalyses.Resultspresentedindicatethat3sRNAsareexclusivetobacteriaoftheBurkholderiacepaciacomplexandhavenohomologuesinotherbacteria,whiletheother21sharehomology,atdifferentextents,tosRNAsofotherbacterialspecies.
