Thestimulationofoocytematurationby1-methyladenineinstarfish,andbyasteroidinfrogs,hasbeenproposedtoinvolveG-protein-coupledreceptors.ToexaminewhetheractivationofreceptorslinkedtoGiorGzwassufficienttocauseoocytematuration,weexpressedmammalianGi-andGz-linkedreceptorsinstarfishandfrogoocytes.Applicationofthecorrespondingagonistscausedmeiosistoresumeinthestarfishbutnotthefrogoocytes.WeconfirmedthatthereceptorswereeffectivelyexpressedinthefrogoocytesbyusingachimericG-protein,Gqi,thatconvertsinputfromGi-andGz-linkedreceptorstoaGqoutputandresultsinacontractionoftheoocyte'spigment.TheseresultsargueagainstGiorGzfunctioningtocausematurationinfrogoocytes.Consistently,maturation-inducingsteroidsdidnotcausepigmentcontractioninfrogoocytesexpressingGqi,andGzproteinwasnotdetectableinfrogoocytes.Forstarfishoocytes,however,ourresultssupporttheconclusionthatGifunctionsin1-methyladeninesignalingandsuggestthepossibilityofusingfrogoocytepigmentcontractionasanassaytoidentifythe1-methyladeninereceptor.Totestthisconcept,wecoexpressedGqiandastarfishadenosinereceptorinfrogoocytesandshowedthatapplyingadenosinecausedpigmentcontraction.
