TheDrosophilatrachealsystemisaninterconnectedtubularrespiratorynetwork,whichisformedbydirectedstereotypicmigrationandfusionofbranches.Cellmigrationandspecificationaredeterminedbycombinatorialsignalingofseveralmorphogenssecretedfromtheectoderm.Wereportthediscoveryofagroupofectodermalcells,markedbyStripe(Sr)expression,thatcoordinatestrachealcellmigrationinthedorsoventralaxis.Sr,anEGRfamilytranscriptionfactor,isknowntoregulatemusclemigration.Inthisstudy,weshowthatSrectodermalcellsalsoprovidesignalsthatareutilizedfortrachealmigration.Thesecuesareseparatedinthetimecourseofembryonicdevelopment.Initially,tendon-precursorcellsareincloseproximitytothetrachealcells,andlater,whentrachealmigrationiscomplete,themusclesdisplacethetracheaandattachtothetendoncells.sr-mutantembryosexhibitdefectsinmigrationofalltrachealbranches.AlthoughtheFGFligandBranchless(Bnl)isexpressedinasubsetoftendon-precursorcellsindependentlyofSr,BnlfunctionscooperativelywithproteinsinducedbySrinattractionoftrachealbranches.
