【 摘 要 】
Pitx2,ahomeodomaintranscriptionfactor,isessentialfornormaldevelopmentofpituitary,eyes,heart,andteeth.Inthedevelopingmousebrain,Pitx2(Rieg,Ptx2,Otlx2,Brx1)mRNAisexpressedindiscreteregionsofthediencephalon,mesencephalon,andrhombencephalon.WhilepriorreportshaveprovidedanoverviewofthetemporalandregionalspecificityofPitx2mRNAexpressioninthebrain,theprecisecelltypesthatexpressPITX2arenotknown.Inthisstudy,weanalyzedPitx2mRNAandPITX2proteinexpressioninindividualcellsofthedevelopinge10.5–e14.5mouseCNSusingmultiplemarkersofcellularproliferationanddifferentiation.WeidentifiedPitx2expressioninnestin-positiveneuralprogenitorsandinpostmitotic,developingneurons.Inthediencephalon,PITX2isexpressedinneuronsofthezonalimitansintrathalamicaandmammillaryregionandinγ-aminobutyricacid(GABA)-producingneuronsofthezonaincerta.Inthemesencephalon,PITX2-labelednucleialsoappearindifferentiatedneurons,someofwhichareGABAergicanddestinedtooccupysuperiorcolliculus.OurresultssuggestthatPITX2expressioninpostmitoticneuronsmaycontributetodevelopmentofGABAergicandotherdifferentiatedneuronalphenotypes.
