【 摘 要 】
Theacrosomereactioninmouseistriggeredbyalong-lastingcalciumsignalingproducedbyachainofopeningsofseveralcalciumchannels,alow-voltage-activated(LVA)calciumchannel,aninositoltrisphosphatereceptor(IP3R),andthestore-operatedcalciumchannelTRP2.Sincematurespermcellsarerefractorytopatchclampexperiments,westudythefunctionalinteractionsamongthosespermcalciumchannelsinspermatogeniccells.Wehavestudiedtheroleofcytosoliccalciuminvoltage-dependentfacilitationoflowvoltage-activatedcalciumchannels.Calciumconcentrationwasmodifiedthroughtheinclusionofthecalciumbuffers,EGTAandBAPTA,intherecordingpipettesolution,andbyadditionofcalciummodulatorslikethapsigarginandthecalciumionophoreA23187.Wedemonstratethatloweringcalciumconcentrationbelowrestinglevelallowstoevidenceavoltage-dependentfacilitation.WealsoshowthatLVAcalciumchannelspresentstrongvoltage-dependentinhibitionbythapsigargin.ThiseffectisindependentofcytosoliccalciumelevationsecondarytocalciumstoredepletionandtotheactivationofTRPchannels.Ourdataevidenceaninterestingfunctionalrelationship,inthiscelltype,betweenLVAchannelsandproteinswhoseactivityisrelatedtocalciumfillingstateoftheendoplasmicreticulum(presumablyTRPchannelsandinositoltriphosphatereceptor).Theserelationshipsmaycontributetotheregulationofcalciumsignalingduringacrosomereactionofmaturespermcell.
