【 摘 要 】

Thepreciseassemblyofanintegratednetworkofbloodvesselsisessentialforthesurvivalofvertebrateembryos.However,theprocessesbywhichprimitiveendothelialcellsformmaturevesselscapableofsupplyingoxygenandnutrientstodevelopingtissuesremainincompletelyunderstood.Here,weproposearoleforRadar,oneofthezebrafishorthologuesofgdf6,inestablishingintegrityofthetrunkvasculatureinzebrafishembryos.Weshowthatradarexpressionisappropriatelyplaced,bothspatiallyandtemporally,toperformsucharole.Transcriptsforradararedetectedinthehypochordandtheprimitivegutendoderm.Thesetissuesintimatelyflankdevelopingaxialvesselsinthetrunkandhavebeenpreviouslyimplicatedintheregulationofvasculardevelopment.Morpholino-basedtargetedgeneknock-downhasgeneratedaRadar-specificloss-of-functionzebrafishmodel.Theseembryosdisplaynormalinitiationofvascularpatterningandcommencementofcirculation.However,byday2ofdevelopment,theintegrityoftheaxialvasculatureiscompromisedwithhemorrhagesandcirculationshort-circuitsthroughoutthedevelopingtrunk.Weshowthatthisaberrantvasculardevelopmentisspecifictoareductionoftheradargeneproduct.TheseresultssuggestthatRadarisinvolvedinasignalingpathwayrequiredforestablishingtheintegrityoftheaxialvesselsduringzebrafishdevelopment.
